1、 Safety precautions for instruments
1. Provide suitable power sockets in the workplace, located away from water environments, and avoid working in explosive or conductive contaminated environments.
2. As the instrument is a hot start device, it needs to be preheated for 20 minutes after starting in summer to evaporate internal water vapor, and the power switch and power cord should be unplugged after each use to ensure safety.
3. One person operates the instrument to avoid damage caused by lack of coordination.
Before operating the instrument, fully prepare the sample before connecting the circuit and turning it on.
5. The pool cover must be closed before operation, and avoid touching the internal metal sheet easily.
2、 Test operation safety
During the experimental operation, breakdown phenomenon is inevitable, which can cause significant damage to the instrument for various reasons. For better maintenance of the instrument, empty samples (only buffer solution) can be used for trial testing during each experiment to verify the safety of the instrument and the purity of the buffer solution used. If any electrical breakdown is found, the operation should be stopped immediately to analyze the cause before conducting the experiment.
Electric explosion phenomenon: If the initial setting parameters are different from the feedback parameters, the feedback values will significantly decrease.
Possible causes of electrical breakdown:
1. Parameter setting error: The relationship between field strength and voltage. The reference data provided by the company is that the field strength needs to be converted.
E=v/c (c is the gap between the electrode cups)
2. The interior of the operation pool should always be kept dry.
If the steps provided by the company require achieving a certain freezing temperature and time, it must be carried out according to the requirements.
4. The electrode cup and buffer solution should be free of impurities. (Mainly from electrolyte contamination)
5. The electrode cup should be washed clean before use, and if used for too long, it needs to be discarded in a timely manner.
6. The interval between two operations should be greater than 1 minute to ensure good heat dissipation of the instrument.
High voltage warning: For practical operation and safety reasons, do not touch any cables or electrode connections during the application of pulses to ECM 399. When connecting or removing cables, check that the system is not in operation or in standby mode.
Some details in the experimental operation:
Electrode cup cleaning process:
Before using electric shock:
1. Use sterilized ultrapure water to repeatedly clean the electrode cup twice (try to touch the gap inside the cup, including the cup lid)
2. Add shock buffer to the electrode cup and clean twice
3. Discard the cleaning solution (discard the liquid) and place it in a super clean bench for UV sterilization for 30 minutes. After the time is up, the experimental operation can be carried out (cover the electrode cup with the lid and place it on ice to wait for the experimental cells)
End of electric shock:
1. Rinse the electrode cup with clean water
2. Rinse 2-3 times with distilled water
3. Soak the electrode cup in 75% alcohol and place it in a beaker or other container for later use (if there is still no need for testing after two weeks, the electrode cup needs to be cleaned every two weeks)
Choose a suitable electrical conversion buffer, preferably a low ion concentration buffer, to reduce the impact on the electrical shock parameters and avoid cup bursting.
The recommended range for sample volume is 50-80 μ l
If there is a cup explosion or spark phenomenon during the electric shock process, there are two reasons:
1. Short circuit caused by high ion content in buffer solution or poor cleaning of electrode cup
2. The sample volume is too small, causing excessive current and resulting in a short circuit
Corresponding solution: Try to eliminate impurities and increase the sample volume to meet the experimental parameter requirements.
